Gaga stars in the action-packed Robert Rodriguez slasher flick as La Chameleón - alongside Amber Heard, Sofia Vergara, Antonio Banderas and Michelle Rodriguez .
The clip shows the 27-year-old 'Born This Way' star dancing alongside a busy highway trying to hitch a ride.
Machete Kills in the second in a series of movies, following on from 2010's Machete - which was previewed as a fake trailer in the cult classic Grindhouse.
The film is set to be released on 13 September 2013.

Lady Gaga recently previewed tracks from her third studio album during a Versace fashion show, with low quality clips of her new music uploaded onto video sharing site, Vine.
There is no release date set for ARTPOP and Gaga has so far revealed little about the album. She has worked with French producer Madeon on the new album, who recently praised working in the studio with the Born This Way star as 'amazing'.
